Modern homonuclear decoupling methods were applied for an easy and unequivocal determination of the acetylation pattern of chitosan. For the first time, diads AD, AA, DA, and AD and deacetylated triads DDD, DDA, ADD, and DAD not depicted by other methods, were firmly assigned The use of 1H-iHOBS facilitated the distribution of acetylated units in randomly deacetylated chitosan samples. Also, the enhancement in sensitivity allowed diads quantitation at a concentration level not possible to address by the well-established 13C-NMR method. Also, we demonstrated that 1H-13C-SAPS-HSQC may be utilized to quantitively assess the properly resolved triads cross-peaks, enhancing chitosan structural information.
